Effects of interferon-γ on cytokine production by endometrial stromal cells

چکیده
To clarify the role of interferon-γ (IFN-γ) in reproduction, we have examined its effects on cytokine production by human endometrial stromal cells (ESC). Concentrations of interleukin (IL)-6, IL-8, monocyte chemoattractant protein1 (MCP-1), and macrophage colony-stimulating factor (M-CSF) in the culture media of normal ESC and an endometrial stromal sarcoma cell line, MaMi, were measured using an enzyme-linked immunosorbent assay. Both non-stimulated ESC and non-stimulated MaMi cells constitutively secrete IL-6, IL-8, MCP-1, and M-CSF. In a dose-dependent manner, IFN-γ increased the concentrations of IL-6, MCP-1, and M-CSF and reduced the concentrations of IL-8 in ESC and MaMi cells. These results suggest that IFN-γ produced by both decidual inflammatory cells and the developing embryo plays a role in the maintenance of early pregnancy by modulating the production of these cytokines by human ESC.

Cytokine Profile in the Endometrium of Normal Fertile and Women with Repeated Implantation Failure
Background: Repeated Implantation Failure (RIF) is one of the most intricate obstacles in assisted reproduction. The cytokine and chemokine composition of uterine cavity seem to play important roles in the implantation process. Objective: To compare the cytokine profile in the endometrium of normal fertile women and those with repeated implantation failure. Methods: After enzymatic digestion of...
متن کاملThe Effect of Beta Interferon on Dendritic Cells and Cytokine Synthesis by CD4+ T Cells
Background: Dendritic cells (DC) are a key regulator of the immune response, and interferon- beta (IFN-β) is considered an immunomodulatory molecule for DC. Objective: The purpose of this study was to evaluate the ability of IFN-β treated DC to induce cytokine secretion by CD4+ T cells. Methods: Dendritic cells were generated from blood monocytes with granulocyte-monocyte colony-stimulating fac...

Background: IFN-g is mostly secreted by activated CD4+ , CD8+ T cells and NK cells. This cytokine has immunomodulatory, anti-cancer and anti-microbial effects and is important for prophylaxis, diagnosis and treatment of chronic infections and cancers. Objective: The purpose of this study was to clone the full cDNA of human IFN-g and express it in CHO cell line. Methods: Lymphocytes from a healt...
متن کاملExpression Pattern of Interferon-γ in Human Leukemic T Cell Lines Following Treatment with Phytoheamagglutinin, phorbol myristate acetate and Lipopolysaccharide
Background: As a T helper type 1 (Th1) derived cytokine, Interferon gamma (IFN-γ) is an important regulator of inflammatory immune responses. Furthermore, IFN-γ plays an essential role in defense against tumors and intracellular pathogens. This study was designed to assess the pattern of IFN-γ production in human leukemic (Jurkat and Molt-4) T cell lines in vitro. Methods: Ju...
